Seminar | Dynamics of T Lymphocyte Fate Specification Revealed by Tracing Cell Proliferative History
Disentangling cell division from time-dependent cell state transitions is key to understanding lineage-specific differentiation. By adapting a well-established fluorescent dye dilution assay for tracking cell proliferative history to mass cytometry, we followed division and time-dependent protein expression and intracellular signaling in single human naïve T cells in the context of expansion for adoptive transfer therapies. In my talk, I will present our findings on the dynamics of early T cell specification revealed by this method. Such an approach to tracking the fate of labeled cells of interest across time and proliferative history will be generally applicable to studies on cell differentiation going forward.
