Signal overlap in mass cytometry is minimal relative to fluorescence-based cytometry in which spectral overlap may be as high as 20% or more of a primary signal. Nevertheless, even when interference is low, a strong signal in one mass channel may still cause background signal in another. Such signal overlap must be taken into consideration when determining gate boundaries for the mass channel receiving undesired signal. This presentation will cover the sources of mass cytometry signal interference, the application of controls to understand how signal interference can impact signal resolution and general principles for optimal panel design.
